Title: Nanopore Tools to Investigate RNA Modifications

Abstract:

Long-read nanopore sequencing has enabled new science to be discovered in DNA and RNA biochemistry. In particular, the ability to detect RNA modifications such as pseudouridine, 5-methylcytidine, and 2’-O-methylated ribonucleotides permits the rapid evaluation of how these modifications change under conditions of cellular stress. We focus on changes in ribosomal RNA modifications because rRNA is the most abundant and long-lived type of RNA in cells, and we believe it can be indicator of cellular health. Examples in this lecture will include (a) the impact of oxidative stress on the mammalian ribosome, (b) an investigation of antibiotic resistance due to mutations and modification changes in bacterial ribosomes exposed to antibiotics, and (c) a study of mRNA vaccine synthesis in which sequence dependent behavior of an RNA polymerase was observed.
